Abstract

Plectranthus amboinicus, a medicinal plant known for its rich content of bioactive compound. However, its high levels of secondary metabolites pose challenges for DNA extraction often resulting in low DNA yield and purity. These limitations can interfere with molecular studies such as PCR and sequencing. Traditional DNA extraction methods like CTAB are effective but labor-intensive and time consuming. As alternatives, rapid DNA extraction methods such as FTA cards and Transdirect Plant Tissue PCR Kit. This study aimed to evaluate and optimize DNA extraction protocols for P. amboinicus using both FTA cards and a Transdirect Plant Tissue PCR kit, addressing in order to overcome the common difficulties encountered with this species.
